For me, it's:
1) Fallout 3 - Perfect Life from E3 2008. http://www.gametrailers.com/player/36201.html
2) Halo 3 - Announcement from E3 2006. http://www.gametrailers.com/player/10454.html
3) Bioshock - Announcement from X06. http://www.gametrailers.com/player/10454.html
Honorable mentions:
FFXIII Announcement from E3 2008 (I don't like the FF series at all, but the trailer's pretty sick) - http://www.gametrailers.com/player/36267.html
Gears of War - Mad World - http://www.gametrailers.com/player/14418.html
The one from Fallout 3, although the game's not out yet, got me from not wanting the game at all to now having it as my most anticipated. The amazing combination of 50's themed TV/commercials along with the song "Dear Hearts and Gentle People" by Bob Crosby and the Bobcats is incredible. The whole theme of the game is incredible (as were the themes of the previous Fallouts of course) and the combination of the two into the trailer is great.
Halo 3's storyline is what pulls me into it the most. I read the first three books when I was younger and I've been hooked since then. Seeing how there's suddenly some gigantic Forerunner structure hidden in the Earth for the first time was incredible. Plus, Martin O'Donnell's incredible music in the backround just threw it over the top. He's done incredible work with the series and proved it right there.
Bioshock's trailer got me for a variety of reasons. Andrew Ryan's (and whoever plays him) incredible speech and voice acting on top of a view of how vast Rapture is...along with the incredibly gory (and somewhat disturbing) scene with the Big Daddy just made that trailer incredible.
And remember, this isn't necessarily just posting trailers of your favorite games, but what games you think have the best trailers.
