Obviously not. GPS needs line of sight and airplane mode turns off ALL transmitting devices on the phone...

If I turn airplane mode on but leave wifi on then the GPS works, but if wifi is off it doesn't work (on my iPhone 4s).

Hmm, I think some phones use the cell-towers to estimate their position aswell, that would be an issue... but GPS recievers like navigation units in your car should only recieve signals from 4 or more sattelites and estimate their location based on the that...

Assisted GPS just helps the GPS get a position using the cell tower, it's not any less of a gps than any other handheld gps. It's an added feature.
GPS in some cars can manage better when it loses the GPS position because it knows how fast the wheels are turning.

At first I was like "WTF", then I realized that some cars have built-in satnav... That's more rare than supercars here, pretty much everyone uses windshield mounted Garmin / TomToms

Dauntless, a lot of window mounted GPS units have the same time except instead of being directly mounted to the car's diagnostics systems, they simply use an accelerometer. This is helpful, for instance, when you go through a tunnel. The GPS can still track your position and tell you about where you are and how much farther you have (and if you need to take the right or the left in the tunnel should you be in one of those kinds).
