I want to go down there for some Scuba Diving, as for the reef life I do think Key Largo is your best bet from what I've read in some of my dive magazines. Not sure about the depth though, maybe you should Scuba Dive while your down there eh?
Key west has a good nightlife. If you are going to fish i'd recommend taking a trip to the tortugas or somewhere between it i heard the fishing is great there. The upper keys are good for fishing too such as islandmorada and key largo. If u are just in to snorkeling then go to lue key off of big pine, its a preservation area that is absolutely beautiful. Key west, islandmorada, and key largo are really the only places where there is stuff to do. Marathon (where i live) has some of the best charters but there is nothing to do besides that.

Somewhere near 400+ miles just to get from the top of Florida to the Keys.

As ronin said visit totugas. Theres a ferry that runs from the key west to the old US fort and you can go snorkeling off the beach on the island with the fort.
