First you need to learn the basics of football,then play practice and play different formations.I'm from Europe,precisely from Croatia where American football isn't very popular.I've started playing since madden 07 and I can say I play it rather good.
I was just like you,the I just played and played until I got a hang of it.
What you need to know is this:
OFFENSE
1.When you choose actions when you're playing a match,you have 2 screens:
1.Formation screen is where you choose in what lineup does your team start the drive(On offense>Running back set,number of WR and TE)
(On defense>number of LB's,DT's and DE's,CB's)
2.(After you choose your formation)Action screen.This where you choose the action(lol)your Off/Def will execute (On offense>If it is a run,then in which direction does your RB go,If it is a pass,then it shows where your WR's are going-it is called routes because they follow specific routes according to action you have chosen)
Here are list of basic routes:
1.Streak - your WR goes straight up
2.Hitch/hook/5 out - your wide receiver goes 5 yards forward then he comes/turns back and stops
3.Post - your WR goes 10/15 yards up then turns to the inside or outside,illustrated like this:

4.slant - your WR does 2/3 steps forward then quickly cuts to inside or outside

DEFENSE
I'm going to explain defense to you because it's harder to play,and you'll going to need to understand it yourself.Here are basics_
After selecting formation,here are explanation of actions you can choose(more like an symbol legend)
Dark blue,purple,yellow,lightish blue blobs/ballons> defender plays zone.It means that he covers specific zone and won't move from.
Arrow - Defender play rush,which means that he blitzes,attacks quarterback
Red arrow - Defender plays man to man defense which means that he covers the opposing player be covering him/following him.
